How often should a standby generator be exercised?

Many manufacturers recommend a short exercise run weekly or every two weeks. Emergency power systems covered by NFPA 110 have their own weekly inspection and monthly test rules.

Does this work for portable generators?

Yes. Remove the transfer switch, block heater and enclosure items and add any items from the owner manual.
